In pharmaceutical and vaccine manufacturing, sterilizing filtration is not merely a processing step — it is the final critical barrier ensuring product sterility, patient safety, and regulatory compliance.

Recently, Filterfine Việt Nam successfully organized a technical seminar titled:

“Comprehensive Filtration Solutions for Pharmaceutical & Vaccine Manufacturing: From Application to Integrity Testing and Filter Validation”

The seminar gathered more than 50 professionals including QA/QC managers, validation engineers, production specialists, and pharmaceutical manufacturers across Vietnam.

Key Highlights from the Filterfine Vietnam Seminar

Participation from Over 35 Pharmaceutical & Biotechnology Organizations

Attendees included representatives from:

  • Vabiotech
  • Viện Kiểm định Vắc xin
  • Pharbaco
  • Meracine
  • Merap Group

along with numerous GMP-WHO pharmaceutical and vaccine manufacturers.

Three Specialized Technical Sessions

The seminar focused on:

  • Life Science filtration solutions
  • Practical integrity testing training
  • Filter validation services for GMP/FDA/WHO compliance

Direct Technical Discussions with Industry Experts

Experts from Entegris addressed practical challenges involving:

  • Cartridge filter reuse
  • CIP/SIP cleaning procedures
  • Bubble Point & Diffusion Flow testing
  • ASTM F838 validation
  • Endotoxin & TOC control
  • Integrity tester troubleshooting

Technical Q&A Highlights

Cleaning and Reusing Sterilizing Filters

Entegris recommends soaking cartridge filters in 1–2% sodium hydroxide solution to remove organic and inorganic contaminants, followed by thorough rinsing with purified water or WFI at 1.5 bar.

For reusable filters, integrity testing is mandatory to verify that filter retentivity remains unaffected after cleaning and sterilization cycles.

From a GMP perspective, filter lifetime depends on:

  • Differential pressure
  • Fluid chemistry
  • Operating temperature
  • SIP/CIP conditions
  • System design

Typically, continued use is not recommended once differential pressure reaches approximately 2 bar.

Why Reverse Flushing Is Not Recommended

Sterilizing-grade membranes possess a complex 3D gradient pore structure with denser downstream layers.

Particles are not only mechanically trapped but also adsorbed within the membrane matrix. Consequently, reverse flushing rarely removes contaminants effectively and may compromise membrane integrity.

For capsule filters, single-use operation is generally recommended to eliminate cross-contamination risks.

Bacterial Retention Capability

Sterilizing-grade filters are validated using:

Brevundimonas diminuta (ATCC 19146)

This microorganism is recognized for its extremely small size:

  • Diameter: 0.3–0.4 μm
  • Length: 0.6–1.0 μm

Validation follows the internationally recognized:

ASTM F838 standard

GMP-WHO Compliance of Filter COA

To comply with WHO GMP, EU GMP, and FDA expectations, filters undergo comprehensive validation including:

Integrity & Bacterial Retention

  • 100% integrity testing
  • Correlation studies between integrity testing and bacterial retention
  • ASTM F838 validation

Cleanliness Validation

  • USP 788
  • USP 645
  • USP 643

Endotoxin Control

  • USP 85

Biological Safety

  • USP 87
  • USP 88

These validations ensure pharmaceutical-grade safety and regulatory compliance.

Temperature Impact on Integrity Testing

Temperature stability is critical during integrity testing because fluctuations affect:

  • Wetting fluid viscosity
  • Surface tension
  • Gas diffusion behavior
  • Pressure and flow stability

Unstable temperatures may result in:

  • False failures
  • False passes

Industry best practices typically require temperature fluctuation control within ±1°C throughout testing.

Troubleshooting Inlet Pressure Errors

Common issues include:

  • Failure to reach target pressure
  • Unstable pressure readings
  • Pressure hold failure

Recommended troubleshooting steps:

  • Perform system leak testing
  • Verify compressed gas supply pressure
  • Ensure complete membrane wetting
  • Contact qualified instrument engineers if necessary
